Nigel Pearson's side had assured themselves of promotion to the Premier League last weekend and they remain on course to go up as champions thanks to a battling display at the Madejski Stadium.
The Foxes are now seven points clear of second-placed Burnley with four games left, while sixth placed Reading hold the final play-off berth ahead of Brighton and Ipswich on goal difference.
Leicester fell behind in the 16th minute when Reading scored their first goal at home from open play in almost nine-and-a-half hours.
Jordan Obita swung a long free-kick into the box and Alex Pearce guided his header back past goalkeeper Kasper Schmeichel into the bottom corner.
Reading should have had a penalty when Dean Hammond clearly handled as he challenged Pavel Pogrebnyak in the air but referee Simon Hooper waved play on.
That proved a crucial decision as Danny Drinkwater equalised for Leicester in the 33rd minute.
Reading only half-cleared a corner to the midfielder 25 yards out and he lashed home to leave Nigel Adkins' side still waiting for a first home win since January.
Mikele Leigertwood, back from a long-term ankle injury, came on as a second-half substitute for Reading, but his only contribution was to collect two quick bookings to force the hosts to play the last few minutes with 10 men.
